Abstract
Physico-chemical and phytochemical standards are of enormous consequence in assess the quality, purity and authenticity of the herbal drugs. Avalguja khadiradi ghrita is a classical formulation mentioned in Bhaisajya Rathnavali under Kushta adikariya for Shwitra. The Avalguja khadiradi ghrita consist thirteen herbal ingredients and Murchitha ghee. The present study was designed to perform the physicochemical and phytochemical standardization with HPTLC fingerprinting of Avalguja khadiradi ghrita according to standard protocols. Avalguja khadiradi ghrita appeared yellow color, specific odor of ghee, viscous and semi solid in nature. Physicochemical analysis of Avalguja khadiradi ghrita showed Saponification value is 269.28, Acid value is 0.03, Iodine value is 10.738, Refractive index is 1. 46546, Viscosity is 58.08, alkaline pH and Specific gravity is 0.9204. Preliminary phytochemical screening of the extracts in different solvent revealed the presence of carbohydrates, phenolic compounds, flavonoids, alkaloids, proteins, saponins, lipids, sterols and tannins. Total flavonoid and phenolic contents in methanolic extract of Avalguja khadiradi ghrita was found to be 1.4 mg/gm and 9.8 mg/gm respectively. Phenolic contents were measured by folin ciocalteu method with help of UV spectrophotometer. Standard calibration curve was plotted by using galic acid as standard phenolic compound. The physicochemical and phytochemical standards along with TLC and HPTLC fingerprint profiles established substantial data for identification, purification and standardization of Avalguja khadiradi ghrita
